Dorsal-ventralspecificationoftheDrosophilaembryoismediatedbysignalingpathwayswhichhavebeenverywelldescribedingeneticterms.However,littleisknownaboutthephysiologyofDrosophiladevelopment.ByimagingpatternsoffreecytosoliccalciuminDrosophilaembryos,wefoundthatseveralcalciumgradientsaregeneratedalongthedorsal-ventralaxis.Themostpronouncedgradientisformedduringstage5,inwhichcalciumlevelsarehighdorsally.Manipulationofthestage5calciumgradientaffectsspecificationoftheamnioserosa,thedorsal-mostregionoftheembryo.Wefurthershowthatthiscalciumgradientisinhibitedinpipe,Toll,anddorsalmutants,butisunalteredindecapentaplegic(dpp)orpuntmutants,suggestingthatthestage5calciumgradientisformedbyasuppressionofventralcalciumconcentrations.Weconcludethatcalciumplaysaroleinspecificationofthedorsalembryonicregion.
