【 摘 要 】

ConventionalmyosinIIactivityprovidesthemotileforceforaxonoutgrowth,buttoachievedirectionalmovementduringaxonpathwayformation,myosinactivityshouldberegulatedbytheattractiveandrepulsiveguidancecuesthatguideanaxontoitstarget.Here,evidenceforthisregulationisobtainedbyusingaconstitutivelyactiveMyosinLightChainKinase(ctMLCK)toselectivelyelevatemyosinIIactivityinDrosophilaCNSneurons.ExpressionofctMLCKpan-neurallyorinprimarilypCC/MP2neuronscausestheseaxonstocrossthemidlineincorrectly.Thisoccurswithoutalteringcellfatesandissensitivetomutationsintheregulatorylightchains.TheseresultsconfirmtheimportanceofregulatingmyosinIIactivityduringaxonpathwayformation.MutationsinthemidlinerepulsiveligandSlit,oritsreceptorRoundabout,enhancethenumberofctMLCK-inducedcrossovers,butctMLCKexpressionalsopartiallyrescuescommissureformationincommissurelessmutants,whererepulsivesignalsremainhigh.OverexpressionofFrazzled,thereceptorformidlineattractiveNetrins,enhancesctMLCK-dependentcrossovers,butcrossoversaresuppressedwhenFrazzledactivityisreducedbyusingloss-of-functionmutations.TheseresultsconfirmthatproperpathwayformationrequirescarefulregulationofMLCKand/ormyosinIIactivityandsuggestthatregulationoccursindirectresponsetoattractiveandrepulsivecues.
