InDrosophila,thedevelopmentofthemidlinecellsoftheembryonicventralnervecorddependsonthefunctionofthebHLH-PAStranscriptionfactorSingle-minded(Sim).Theexpressiondomainofsim,however,isalsofoundanteriorandposteriortothedevelopingventralcordthroughoutthegermband.Indeed,mutationsinsimwereidentifiedbasedontheircharacteristiccuticlephenotype.Eightabdominalsegments(A1–A8)canbeeasilyseeninthelarvalcuticle,whilethreemorecanbeidentifiedduringembryogenesis.CellslocatedinA8–A10giverisetotheformationofthegenitalimaginaldiscs,andahighlymodifiedA11segmentgivesrisetotheanalpadsthatflanktheanus.simisexpressedinallthesesegmentsandisrequiredfortheformationofboththeanalpadsandthegenitalimaginaldiscs.Anewtemperature-sensitivesimalleleallowedanassessmentofpossiblepostembryonicfunction(s)ofsim.Reductionofsimfunctionbelowa50%thresholdleadstosterileflieswithmarkedbehavioraldeficits.Mostmutantsimflieswereonlyabletowalkincircles.Furtheranalysesindicatedthatthisphenotypeislikelyduetodefectsinthebraincentralcomplex.Thisbrainregion,whichhaspreviouslybeenimplicatedinthecontrolofwalkingbehavior,expresseshighlevelsofnuclearSimproteininthreeclustersofneuronsineachcentralbrainhemisphere.AdditionalSimlocalizationinthemedullaryandlaminarneuronsoftheopticlobesmaycorrelatewiththepresenceofectopicaxonbundlesobservedintheopticlobesofsimmutantflies.
