【 摘 要 】

Developmentofthecartilaginouscapsuleoftheinnerearisdependentoninteractionsbetweenoticepitheliumanditssurroundingperioticmesenchyme.Duringthesetissueinteractions,factorsendogenoustotheoticepitheliuminfluencethedifferentiationoftheunderlyingperioticmesenchymetoformachondrifiedoticcapsule.WereportthelocalizationofSonichedgehog(Shh)proteinandexpressionoftheShhgeneinthetissuesofthedevelopingmouseinnerear.WedemonstrateinculturesofperioticmesenchymethatShhalonecannotinitiateoticcapsulechondrogenesis.However,whenShhisaddedtoculturedperioticmesenchymeeitherincombinationwithoticepitheliumoroticepithelial-derivedfibroblastgrowthfactor(FGF2),asignificantenhancementofchondrogenesisoccurs.AdditionofShhantisenseoligonucleotide(AS)toculturedperioticmesenchymewithaddedoticepitheliumdecreaseslevelsofendogenousShhandsuppressesthechondrogenicresponseofthemesenchymecells,whilesupplementationofShhAS-treatedcultureswithShhrescuesculturesfromchondrogenicinhibition.WedemonstratethatinactivationofShhbytargetedmutationproducesanomaliesinthedevelopinginnerearanditssurroundingcapsule.OurresultssupportaroleforShhasaregulatorofoticcapsuleformationandinnereardevelopmentduringmammalianembryogenesis.
