【 摘 要 】

ThelatentTGF-βbindingproteins(LTBP)arebelievedtocontroltheavailabilityofTGF-βintheextracellularmilieu.TogaininsightintothepotentialrolesofLTBPinearlydevelopment,weisolatedtheXenopusLTBP-1(xLTBP-1)cDNA.ThecDNAencodesaproteinsimilartothemammalianLTBP-1inbothsizeanddomainstructure.Inaddition,wefoundanovellongerspliceisoformofxLTBP.TheRNAsforbothformsofxLTBPdisplayedtemporalregulationandtheshortertranscriptisexpressedmaternally.Bothtranscriptsalsodisplayspatialregulationandarefoundinthedorsalmesodermoftheorganizer.Inanimalcapexperiments,LTBP-1potentiatestheactivityofactivinandnodal.TheactivityofLTBP-1didnotappeartorequirecovalentassociationwithactivinastheadditionofmediumcontainingactivinandLTBP-1toanimalcapsenhancedtheactivineffect.TheseresultsindicatethatLTBP-1maybepartoftheregulatorysystemthatestablishesthethresholdofmorphogenactivityforactivinsandnodalsinthedorsalsideoftheembryoduringgastrulation.
