【 摘 要 】
CD9isamembraneproteinbelongingtothetetraspaninfamily.DespiteCD9'sbroadtissuedistribution,theonlyabnormalityobservedinCD9-deficientmicewasinfertilityoffemales,whichwasresponsibleforadefectinthesperm–eggfusionprocess.However,thefunctionofCD9insperm–eggfusionisnotclearatallbecausethetechniquetoanalyzetheactivityofmoleculesinsperm–eggfusionhasnotbeenestablished.WedemonstratedthattheexogenousmouseCD9,expressedbypolyadenylatedmRNAinjectionatthegerminal-vesiclestageoocytes,waspreciselylocalizedtotheeggplasmamembrane,andtheexpressionreversedtheinfertilityofCD9−/−eggs.Then,twoothertetraspanins,humanCD9andmouseCD81,overexpressedwiththistechniqueonCD9−/−eggsrestoredthefertilizationrateupto∼90and∼50%againstthatofwildtypeeggs,respectively.Moreover,inthepresenceofananti-mouseCD9mAb,whichblockssperm–eggfusion,expressionofhumanCD9ormouseCD81oneggsalsorescuedthefusibility.TheseresultssuggestedthathumanCD9playsacrucialroleinhumanfertilization,andmouseCD81hasthepotentialtocompensateforCD9functioninsperm–eggfusion.Inaddition,thepolyadenylatedmRNAinjectioniseffectiveformolecularanalysisofsperm–eggfusion.
