Thesecondmessengercyclicadenosine5′monophosphate(cAMP)hasbeenimplicatedincontrollingmeioticmaturation.Todate,therehavebeennodirectmeasurementsofcAMPinlivingmammalianoocytes.Here,wehaveusedthefluorescentlylabelledcAMP-dependentproteinkinaseA(PKA),FlCRhR,tomonitorcAMPinmouseoocytes.Incumulus-enclosedoocytes,follicle-stimulatinghormone(FSH)stimulatedanincreaseintheoocyte[cAMP]thatwaspreventedbyusingthegapjunctioninhibitor,carbenoxolone.TheFSH-inducedincreaseinoocyte[cAMP]wassuppressedinatime-dependentmannerbypriorexposuretoATP,whileepidermalgrowthfactorhadnoeffectonbasalorstimulatedlevelsofcAMP.Finally,usingconfocalmicroscopy,weshowthattheregulatoryandcatalyticsubunitsofthemicroinjectedPKAaredistributedinapunctatemannerwithastrongeraccumulationintheperinuclearregion.Onanincreasein[cAMP],inresponsetophosphodiesteraseinhibitionorFSH,thecatalyticsubunitdiffusedthroughoutthecytoplasmandgerminalvesicle,whiletheregulatorysubunitremainedanchored.TheseexperimentsshowthatincreasesincAMPinovariansomaticcellsarecommunicatedviagapjunctionstotheoocyte,whereitcanleadtoaredistributionofthecatalyticsubunitofPKA.
