OneofthemostsignificantmorphogeniceventsinthedevelopmentofDrosophilamelanogasteristheelongationofimaginaldiscsduringpupariumformation.WehaveshownthatthismacroscopiceventisaccompaniedbytheformationofGolgistacksfromsmallGolgilarvalclustersofvesiclesandtubulesthatarepresentpriortotheonsetofdiscelongation.Wehaveshownthattheflysteroidhormone20-hydroxyecdysonetriggersboththeelongationitselfandtheformationofGolgistacks(V.Kondylis,S.E.Goulding,J.C.Dunne,andC.Rabouille,2001,Mol.Biol.Cell,12,2308).UsingmRNAinsituhybridisation,weshowherethatecdysonetriggerstheupregulationofasubsetofgenesencodingGolgi-relatedproteins(suchasdnsf1,dsec23,dsed5,anddrab1)anddownregulatestheexpressionofothers(suchasdergic53,dβ'COP,anddrab6).WeshowthatthetranscriptionfactorBroad-complex,itselfan“earlyâ€ecdysonetarget,mediatesthisregulation.Andweshowthattheecdysone-independentupregulationofdnsf1anddsnappriortotheecdysonepeakleadstoaprecociousformationoflargeGolgistacks.Theecdysone-triggeredbiogenesisofGolgistacksattheonsetofimaginaldiscelongationofferstheexcitingpossibilityofadvancingourunderstandingoftherelationshipbetweengeneexpressionandorganellebiogenesis.
