Onemajordifferencebetweensimplemetazoanssuchascnidariansandallthebilateriananimalsisthoughttoinvolvetheinventionofmesoderm.Thetermsdiploblastsandtriploblastsaretherefore,oftenusedtogroupprebilaterianandbilateriananimals,respectively.However,jellyfishcontainwelldevelopedstriatedandsmoothmuscletissuesthatderivefromtheentocodon,amesoderm-liketissueformedduringmedusadevelopment.Weinvestigatedthehypothesis,thattheentocodoncouldbehomologoustothethirdgermlayerofbilateriansbyanalyzingthestructuresandexpressionpatternsofthehomologuesofBrachyury,Mef2,andSnailinthejellyfishPodocorynecarnea.TheseareregulatorygenesfromtheT-box,MADS-boxandzincfingerfamiliesknowntoplayimportantrolesinbilaterianmesodermpatterningandmuscledifferentiation.ThesequenceandexpressiondatademonstratethatthegenesarestructurallyandfunctionallyconservedandevenmoresimilartohumansorotherdeuterostomesthantoprotostomemodelorganismssuchasDrosophilaorCaenorhabditiselegans.Basedonthesedataweconcludethatthecommonancestorofthecnidariansandbilateriansnotonlysharedgenesthatplayaroleinregulatingmyogenesisbutalreadyusedthemtodevelopanddifferentiatemusclesystemssimilartothoseoftriploblasts.
