【 摘 要 】

Completesexualdevelopmentisnoteasilyamenabletoexperimentationinhydra.Therefore,theanalysisofgenefunctionandgeneregulationrequirestheintroductionofexogenousDNAinalargenumberofcellsofthehydrapolypsandthesignificantexpressionofreporterconstructsinthesecells.WepresentheretheprocedurewherebywecoupledDNAinjectionintothegastriccavitytoelectroporationofthewholeanimalinordertoefficientlytransfecthydrapolyps.WecoulddetectGFPfluorescenceinbothendodermalandectodermalcelllayersofliveanimalsandinepithelialaswellasinterstitialcelltypesofdissociatedhydra.Inaddition,wecouldconfirmGFPproteinexpressionbyshowingcolocalisationbetweenGFPfluorescenceandanti-GFPimmunofluorescence.Finally,whenaFLAGepitopewasinsertedin-framewiththeGFPcodingsequence,GFPfluorescencealsocolocalisedwithanti-FLAGimmunofluorescence.ThisGFPexpressioninhydracellswasdirectedbyvariouspromoters,eitherhomologous,likethehydrahomeoboxcnox-2genepromoter,orheterologous,likethetwonematoderibosomalproteinS5andL28genepromoters,andthechickenβ-actingenepromoter.Thisstrategyprovidesnewtoolsfordissectingdevelopmentalmolecularmechanismsinhydra;morespecifically,thegeneticregulationsthattakeplaceinendodermalcellsatthetimebuddingorregenerationisinitiated.
