【 摘 要 】
Duringcorticaldevelopment,differentcellpopulationsariseinthebasaltelencephalonandsubsequentlymigratetangentiallytotheneocortex.However,itisnotclearwhetherthesecorticalcellsaregeneratedinthelateralganglioniceminence(LGE),themedialganglioniceminence(MGE),orboth.Inthisstudy,wehavegeneratedathree-dimensionalreconstructiontostudythemorphologicalformationofthetwoganglioniceminencesandtheinterganglionicsulcus.Asaresult,wehavedemonstratedtheimportanceofthedevelopmentofthesestructuresforthistangentialmigrationtotheneocortex.WehavealsousedthetracersDiIandBDAinmultipleexperimentalparadigms(wholeembryoculture,inuteroinjections,andbrainslicecultures)toanalyzetheroutesofcellmigrationandtodemonstratetherolesofbotheminencesinthedevelopmentofthecerebralcortex.Theseresultsarefurtherstrengthened,confirmingtheimportanceoftheMGEinthismigrationanddemonstratingtheearlygenerationoftangentialmigratorycellsintheLGEearlyindevelopment.Finally,weshowthatthecalcium-bindingproteinCalretininisexpressedinsomeofthesetangentiallymigratingcells.Moreover,wedescribethespatiotemporalsequenceofGABA,Calbindin,andCalretininexpression,showingthatthesethreemarkersareexpressedinthecorticalneuroepitheliumoverseveralembryonicdays,suggestingthatthecellsmigratingtangentiallyformaheterogeneouspopulation.
