TheDictyosteliumproteinAmpA(adhesionmodulationproteinA)isencodedbythegeneoriginallyidentifiedbytheD11cDNAclone.AmpAcontainsrepeateddomainshomologoustoavarietyofproteinsthatinfluencecelladhesion.Theproteinaccumulatesduringdevelopment,reachingamaximallevelatthefingerstage.MuchoftheAmpAproteinisfoundextracellularlyduringdevelopment,andinculminants,AmpAisfoundinassociationwithanterior-likecells.CharacterizationofanampA−straingeneratedbygenereplacementrevealsasignificantincreaseincell–cellclumpingwhencellsarestarvedinnonnutrientbuffersuspensions.DevelopingampA−cellsarealsomoreadhesivetotheunderlyingsubstrateandaredelayedindevelopmentalprogression,withtheseverityofthedelayincreasingascellsaregrowninthepresenceofbacteriaorontissueculturedishesratherthaninsuspensionculture.ReintroductionoftheampAgenerescuesthedevelopmentaldefectsofampA−cells;however,expressionofadditionalcopiesofthegeneinwild-typecellsresultsinmoreseveredevelopmentaldelaysanddecreasedclumpinginsuspensionculture.WeproposethattheAmpAproteinfunctionsasananti-adhesivetolimitcell–cellandcell–substrateadhesionduringdevelopmentandthusfacilitatescellmigrationduringmorphogenesis.
