【 摘 要 】

Transientelevationsofintracellularcalcium(calciumtransients)playcriticalrolesinmanydevelopmentalprocesses,includingdifferentiation.Althoughthefactorsthatregulatecalciumtransientsarenotclearlydefined,calciuminfluxmaybecontrolledbymoleculesinteractingwithcalciumchannels,includingchannelregulatorysubunits.Here,wedescribethechickγ4regulatorysubunit(CACNG4),thefirstsuchsubunittobecharacterizedinearlydevelopment.CACNG4isexpressedearlyinthecranialneuralplate,andlaterinthecranialanddorsalrootganglia;importantly,thetimingofthislaterexpressioncorrelatespreciselywiththeonsetofneuronaldifferentiation.CACNG4expressionisalsoobservedinnonneuronaltissuesundergoingdifferentiation,specificallythemyotomeandasubpopulationofdifferentiatingmyoblastsinthelimbbud.Finally,withinthedistalcranialganglia,weshowthatCACNG4isexpressedinplacode-derivedcells(prospectiveneurons),butalso,surprisingly,inneuralcrest-derivedcells,previouslyshowntoformonlygliainthislocation;contrarytothesepreviousresults,wefindthatneuralcrestcellscanformneuronsinthedistalganglia.GiventheproposedroleofCACNG4inmodulatingcalciumchannelsanditsexpressionindifferentiatingcells,wesuggestthatCACNG4maypromotedifferentiationviaregulationofintracellularcalciumlevels.
