【 摘 要 】
Jointformation,theonsetofwhichischaracterizedbythesegmentationofcontinuousskeletalrudimentsintotwoormoreseparateelements,isafundamentalaspectoflimbpatternformation,playingacriticalroleindeterminingthesize,shape,andnumberofindividualskeletalelements.Jointformationisinitiatedbyconversionofdifferentiatedchondrocytesatsitesofpresumptivejointsintodenselypackednonchondrogeniccellsofthejointinterzone.ThisconversionisaccompaniedbylossofAlcianblue-stainingcartilagematrixanddownregulationofcartilage-specificgeneexpression.Here,wereportthatCux1,whichencodesatranscriptionfactorcontainingahomeodomainandotherDNA-bindingmotifs,ishighlyexpressedatallofthediscretesitesofincipientjointformationinthedevelopinglimbconcomitantwithconversionofdifferentiatedchondrocytesintointerzonetissue.Moreover,differentiatedlimbchondrocytesinmicromassculturesinfectedwithaCux1retroviralexpressionvectorareconvertedintononchondrogeniccellswhichexhibitlossofAlcianbluecartilagematrixanddownregulationofcartilage-specificgeneexpressionasoccursattheonsetofnormaljointformation.TheseresultssuggestthatCux1isinvolvedinregulatingtheonsetofjointformationbyfacilitatingconversionofchondrocytesintononchondrogeniccellsoftheinterzone.
