【 摘 要 】

Dynamicepigeneticmodificationofthegenomeoccursduringearlydevelopmentofthemouse.Activedemethylationofthepaternalgenomeoccursinthezygote,followedbypassivedemethylationduringcleavagestages,anddenovomethylation,whichisthoughttohappenafterimplantation.Wehaveinvestigatedtheseprocessesbyusingindirectimmunofluoresencewithanantibodyto5-methylcytosine.Incontrasttopreviouswork,weshowthatdemethylationofthemalepronucleusiscompletedwithin4hoffertilisation.Thisactivityisintricatelylinkedwithandnotseparablefrompronucleusformation.Inconditionspermissiveforpolyspermy,uptofivemalepronucleiunderwentdemethylationinthesameoocyte.PaternaldemethylationinfertilisedoocytesdeficientforMBD2,theonlycandidatedemethylase,occurrednormally.Passivelossofmethylationoccurredinastepwisefashionuptothemorulaestagewithoutanyevidenceofspatialcompartmentalisation.Denovomethylationwasobservedspecificallyintheinnercellmass(ICM)butnotinthetrophectodermoftheblastocystandhencemayhaveanimportantroleinearlylineagespecification.Thisisthefirstcompleteanddetailedanalysisoftheepigeneticreprogrammingcycleduringpreimplantationdevelopment.Thethreephasesofmethylationreprogrammingmayhaverolesinimprinting,thecontrolofgeneexpression,andtheestablishmentofnucleartotipotency.
