Thegiantproteinisashort-rangetranscriptionalrepressorthatrefinestheexpressionpatternofgapandpair-rulegenesintheDrosophilablastodermembryo.Short-rangerepressorsincludingknirps,Krüppel,andsnailutilizetheCtBPcofactorforrepression,butitisnotknownwhetherafunctionalinteractionwithCtBPisageneralpropertyofallshort-rangerepressors.WestudiedgiantrepressionactivityinaCtBPmutantandfindthatthiscofactorisrequiredforgiantrepressionofsome,butnotall,genes.Whiletargetsofgiantsuchastheeven-skippedstripe2enhancerandasyntheticlacZreportershowclearderepressionintheCtBPmutant,anothergianttarget,thehunchbackgene,isexpressednormally.AmorecomplexsituationisseenwithregulationoftheKrüppelgene,inwhichoneenhancerisrepressedbygiantinaCtBP-dependentmanner,whileanotherisrepressedinaCtBP-independentmanner.TheseresultsdemonstratethatgiantcanrepressbothviaCtBP-dependentandCtBP-independentpathways,andthatpromotercontextiscriticalfordetermininggiant-CtBPfunctionalinteraction.Toinitiatemechanisticstudiesofthegiantrepressionactivity,wehaveidentifiedaminimalrepressiondomainwithingiantthatencompassesresidues89–205,includinganevolutionarilyconservedregionbearingaputativeCtBPbindingmotif.
