【 摘 要 】
Theoriginofresident(noninflammatory)macrophagesinvertebratetissuesisstillpoorlyunderstood.Inthezebrafishembryo,werecentlydescribedaspecificlineageofearlymacrophagesthatdifferentiateintheyolksacbeforetheonsetofbloodcirculation.Wenowshowthattheseearlymacrophagesspreadinthewholecephalicmesenchyme,andfromthereinvadeepithelialtissues:epidermis,retina,andbrain—especiallytheoptictectum.Inthepanthermutant,whichlacksafunctionalfms(M-CSFreceptor)gene,earlymacrophagesdifferentiateandbehaveapparentlynormallyintheyolksac,butthenfailtoinvadeembryonictissues.Ourvideorecordingsthendocumentforthefirsttimethebehaviorofmacrophagesintheinvadedtissues,revealingthestrikingpropensityofearlymacrophagesinepidermisandbraintowanderrestlesslyamongepithelialcells.Thisunexpectedbehaviorsuggeststhattissuemacrophagesmaybeconstantly“patrollingâ€forimmuneandpossiblyalsodevelopmentalandtrophicsurveillance.At60hpost-fertilization,allmacrophagesinthebrainandretinaundergoaspecificphenotypictransformation,into“early(amoeboid)microgliaâ€:theybecomemorehighlyendocytic,theydown-regulatetheL-plastingene,andabruptlystartexpressinghighlevelsofapolipoproteinE,awell-knownneurotrophiclipidcarrier.
