【 摘 要 】

CNSgliahaveintegralrolesindirectingaxonmigrationofbothvertebratesandinsects.Incontrast,verylittleisknownabouttherolesofPNSgliainaxonalpathfinding.InvertebratesandDrosophila,anatomicalevidenceshowsthatperipheralgliaprefigurethetransitionzonesthroughwhichaxonsmigrateintoandoutoftheCNS.Therefore,peripheralgliacouldguideaxonsatthetransitionzone.WeusedtheDrosophilamodelsystemtotestthishypothesisbyablatingperipheralgliaearlyinembryonicneurodevelopmentviatargetedoverexpressionofcelldeathgenesgrimandced-3.Theeffectsofperipheralgliallossonsensoryandmotorneurondevelopmentwereanalyzed.MotoraxonsinitiallyexittheCNSinabnormalpatternsintheabsenceofperipheralglia.However,theymustuseothercueswithintheperipherytofindtheircorrecttargetmusclessinceearlypathfindingerrorsarelargelyovercome.Whenperipheralgliaarelost,sensoryaxonsshowdisruptedmigrationastheytravelcentrally.Thisisnotaresultofmotorneurondefects,asdeterminedbymotor/sensorydouble-labelingexperiments.WeconcludethatperipheralgliaprefiguretheCNS/PNStransitionzoneandguideaxonsastheytraversethisregion.
