【 摘 要 】

IndividualsomaticmusclesandheartprogenitorsarespecifiedatdefinedpositionswithinthemesodermallayerofDrosophila.Theexpressionofthehomeoboxgeneeven-skipped(eve)identifiesonespecificsubsetofcellsinthedorsalmesoderm,whichgiverisetoparticularpericardialcellsanddorsalbodywallmuscles.Geneticanalysishasshownthattheinductionofeveinthesecellsinvolvesthecombinedactivitiesofgenesencodingmesoderm-intrinsicfactors,suchasTinman(Tin),andspatiallyrestrictedsignalingactivitiesthatarelargelyderivedfromtheectoderm,particularlythoseencodedbywingless(wg)anddecapentaplegic(dpp).HereweshowthataDpp-activatedSmadprotein,phosphorylatedMad,iscolocalizedineve-expressingcellsduringanextendeddevelopmentalperiod.WedemonstratefurtherthatamesodermallyactiveenhancerofevecontainsseveralSmadandTinbindingsitesthatareessentialforenhanceractivityinvivo.ThisenhanceralsocontainsanumberofbindingsitesfortheWg-effectorPangolin(Pan/Lef-1),whicharerequiredforfulllevelsofenhanceractivity.However,wefindthattheirmainfunctionistopreventectopicenhanceractivityinthedorsalmesoderm.Thissuggeststhat,intheabsenceofWgsignaling,PanbindingservestoabrogatethesynergisticactivitiesofSmadsandTinineveactivationwhile,incellsthatreceiveWgsignals,Panisconvertedintoacoactivatorthatpromoteseveinduction.Together,thesedatashowthattheeveenhancerintegratesseveralregulatorypathwaysviathecombinatorialbindingofthemesoderm-intrinsicregulatorTinandtheeffectorsoftheDppandWgsignals.
