Withindevelopingovariesofmanyinsects,somedevelopingfolliclesoroocytesusuallydegenerate(follicularatresiaoroosorption),whiletheothersmaycontinuetogrowtomaturity,thusmaintainingthebalancebetweenthenumberofeggsandreproductivecircumstancessuchasavailablenutrients.Tohelpclarifythephenomenonoffollicularatresiaduringovariandevelopment,weexaminedcysteineproteinasesstoredinmosquitoCulexpipienspallensovaries.First,analysisusingsynthesizedsubstratesshowedthatcathepsinB-andL-likeproteinasesgraduallyaccumulatedinthedevelopingovariesafterabloodmeal,whichrequiredmorethan10minofpreincubationunderacidicconditionstoreachtheirmaximumactivities.However,homogenatesofdegeneratingfollicles3daysafterfeedingshowedproteolyticactivitieswithoutacidtreatment,suggestingthattheproteinaseshadalreadybeenactivated,whiletheextractofnormallydevelopingfolliclescollectedfromthesameovariesrequiredmorethan10minofacidpreincubationtoreachtheoptimumactivities,suggestingthattheenzymesremainedasinactiveforms.Chemicalandimmunohistochemicalanalysesshowedthatmoreproteinasesarelocatedinthecytoplasm,ratherthanbeingassociatedwithyolkgranules.Ovarianproteinases,whicharebelievedtobecomeactivatedattheonsetofembryogenesis,shouldalsobeactivatedduringoogenesis,presumablytoenhanceoosorption.
