FertilizationofstarfisheggsduringmeiosisresultsinrapidprogressiontoembryogenesisassoonasmeiosisIIiscompleted.Unfertilizedeggscompletemeiosisandarrestinpostmeioticinterphaseforan,untilnow,indeterminatetime.Iftheyremainunfertilized,thematurepostmeioticeggsultimatelydie.Theaimofthisstudyistocharacterizethemechanismofdeathinpostmeioticunfertilizedstarfisheggs.Wereportthat,intwospeciesofstarfish,intheabsenceoffertilization,postmeioticinterphasearrestpersistsfor16–20h,afterwhichtimethecellssynchronouslyandrapidlydie.Dyingeggsextrudemembraneblebs,undergocytoplasmiccontractionanddarkening,andfragmentintovesiclesinamannerreminiscentofapoptoticcells.TheDNAofdyingeggsiscondensed,fragmented,andlabeledbytheTUNELassay.Takentogether,thesedatasuggestthatthedefaultfateofpostmeioticstarfisheggs,liketheirmammaliancounterparts,isdeathbyapoptosis.WefurtherreportthattheonsetandexecutionofapoptosisinthissystemisdependentonongoingproteinsynthesisandisinhibitedbyariseinintracellularCa2+,anessentialcomponentofthefertilizationsignalingpathway.Weproposestarfisheggsasausefulmodeltostudydevelopmentallyregulatedapoptosis.
