Programmedcelldeathisaprominentfeatureofembryonicdevelopmentandisessentialinmatchingthenumberofneuronstothetargettissuesthatareinnervated.Althoughadecreaseinneuronalnumberwhichcoincideswithperipheralsynaptogenesishasbeenwelldocumentedintheavianciliaryganglion,ithasnotbeenclearwhethercelldeathalsooccursearlier.WeobservedTUNEL-positiveneuronsasearlyasstage24,withalargepeakatstage29.Thiscelldeathatstage29wasfollowedbyastatisticallysignificant(P<0.0001)decreaseintotalneuronnumberatstage31.Thetotalnumberofneuronswasrecoveredbystage33/34.Thissuggestedthatdyingneuronswerereplacedbynewneurons.Thisreplacementprocessdidnotinvolveproliferationbecausebromodeoxyuridineappliedatstages29and31wasunabletolabelneuronsharvestedatstage33/34.Thepeakofcelldeathatstage29wasincreased2.3-foldbyremovaloftheopticvesicleandwasreducedby50%whenchCNTFwasoverexpressed.Takentogether,theseresultssuggestthattheregulationofneuronnumberintheciliaryganglionisadynamicprocessinvolvingbothcelldeathandneuralreplacementfrompostmitoticprecursorspriortodifferentiationandinnervationoftargettissues.
