【 摘 要 】

Thethymidineanalog5-bromodeoxyuridine(BUdR)iswellknownforitsmutageniceffectswhenincorporatedinDNA.Similarly,BUdRhasbeenreportedbyseveralworkerstoinhibitcytodifferentiationwhenappliedduringthecellproliferativephase.Wehaveobservedthatrelativelylowlevels(ca10−6M)ofBUdR,IUdR,ClUdR,andBCdRallcauseagradualchangeincellmorphology,andinhibitthefusionofcellstoformsyncytiainmuscleclones,butdonotinterferewithproliferationofmononuclearcells.Theinhibitionofdifferentiationcanbereversedbyfurthercelldivisionintheabsenceoftheanalog.Afterseveraldays,thisresultsinclonalcultureswhichareindistinguishablefromuntreatedcontrolsbothintermsoftotalnumberofclonesandoftheproportionofcloneswhicharemyogenic;thusthedrugsarenotactingeitherbycellselectionormutagenesis.BUdRisincorporatedintoDNAinplaceofthymine,itseffectondifferentiationcanbecompetitivelypreventedbysimultaneousinclusioninthemediumofTdRorcompoundsreadilyconvertibletoTdR,andithasnodetectableeffectonpostmitoticcells(musclefibers).AlthoughthissuggeststhatBUdRexertsitseffectondifferentiationfromasiteintheDNA,aneffectoncellmetabolismatsomeotherlevelcannotberuledout.
