Thisreviewcriticallyexaminesthehypothesisthatmolecularchaperonesandprotein‐foldingcatalystsfromprokaryotesandeukaryotescanbesecretedbycellsandfunctionasintercellularsignals,principallybutnotexclusively,forleukocytes.Agrowingnumberofmolecularchaperoneshavebeenreportedtofunctionasligandsforselectedreceptorsand/orreceptorsforspecificligands.Molecularchaperonesinitiallyappearedtoactprimarilyasstimulatorysignalsforleukocytesandthus,wereseenasproinflammatorymediators.However,evidenceisnowemergingthatmolecularchaperonescanhaveanti‐inflammatoryactionsor,dependingontheproteinandconcentration,anti‐andproinflammatoryfunctions.Recastingtheoriginalhypothesis,weproposethatmolecularchaperonesandprotein‐foldingcatalystsare“moonlighting”proteinsthatfunctionashomeostaticimmuneregulatorsbutmayalsoundercertaincircumstances,contributetotissuepathology.Oneofthekeyissuesinthefieldofmolecularchaperonebiologyrelatestotheroleofmicrobialcontaminantsintheirsignalingactivity;thistoowillbeevaluatedcritically.Themostfascinatingaspectofmolecularchaperonesprobablyrelatestoevidencefortheirtherapeuticpotentialinhumandisease,andongoingstudiesareevaluatingthispotentialinarangeofclinicalsettings...
