TheC‐typelectinSIGNR3isamousehomologueofhumanDC‐SIGN,whichsharescarbohydrate‐bindingspecificitywithhumanDC‐SIGN.However,theexpressionprofileofSIGNR3islargelyunknown.ToexaminetheexpressionofSIGNR3inimmunecells,wegeneratedSIGNR3‐specificmAbandinvestigatedSIGNR3expressioninvivo.SIGNR3wasexpressedonafractionofMHCII+DCsandMφsinthedermisandCD115+Ly6Cint‐lowmonocytesinthebloodandBM.IntheLNs,SIGNR3+cellslocalizedadjacenttoPNAd+HEV‐likevessels.TheywerealsofoundininterfollicularregionsinsLNsbutnotmLNs.ThoseSIGNR3+cellsexpressedCD11bandvariablelevelsofCD11candMHCII.AsinLNs,SIGNR3wasexpressedonalargeproportionoftheCD11b+CD11cint‐highcellsinthespleen.Inthelung,SIGNR3+cellsbelongedtotheCD11b+CD11cintpopulation,andMφsintheairwayandlungfaintlyexpressedSIGNR3.WhenPKH67‐labeledCD115+Ly6ChighBMmonocytesweretransferredintonormalrecipients,theyup‐regulatedSIGNR3expressionalongwiththedecreaseinLy6CexpressionduringthecirculationanduponarrivalattheperipheralLNsthroughHEV.Inaddition,CD11bhighLy6ChighmonocytesthatenteredsLNsdifferentiatedintoCD11b+DCsinacoupleofdays,whereasthoseinthespleen,mLNs,andlungdifferentiatedintoCD11cintmonocyticcells.TheseresultssuggestthatSIGNR3isanewdifferentiationmarkerformyeloidmononuclearcellsandindicatethatsomeDCs,especiallyinthesLNs,arepossiblyreplenishedbyLy6Chighmonocytes...
