Herein,weinvestigatedtheinvolvementofthe5‐LO‐derivedlipidmediatorLTB4inγδTcellmigration.Wheninjectedintothei.pl.spaceofC57BL/6mice,LTB4triggeredγδTlymphocytemobilizationinvivo,aphenomenonalsoobservedininvitrochemotaxisassays.Thei.pl.injectionofEscherichiacoliendotoxin(LPS)triggeredincreasedlevelsofLTB4inpleuralcavities.TheinvivoinhibitionofLTB4biosynthesisbythe5‐LOinhibitorzileutonortheFLAPinhibitorMK886attenuatedLPS‐inducedγδTcellaccumulationintopleuralcavities.Accordingly,5‐LOKOmicefailedtorecruitγδTcellsintotheinflammatorysiteafteri.pl.LPS.Antagonistsofthehigh‐affinityLTB4receptorBLT1,CP105,696,andLY292476alsoattenuatedLPS‐inducedγδTcellaccumulationinpleuralcavitiesaswellasinvitrochemotaxistowardpleuralwashesobtainedfromLPS‐simulatedmice.LTB4/BLT1alsoaccountedforγδTcellmigrationinducedbyi.pl.administrationofMycobacteriumbovisBCGorantigeninsensitizedmice.BLT1wasexpressedonnaïve,residentaswellasLPS‐recruitedγδTcells.IsolatedγδTcellswerefoundtoundergoF‐actincytoskeletonreorganizationwhenincubatedwithLTB4invitro,confirmingthatγδTlymphocytescanresponddirectlytoLTB4.InadditiontoitsdirecteffectonγδTcells,LTB4triggeredtheiraccumulationindirectly,viamodulationofCCL2productioninmousepleuralcavities.ThesedatashowthatγδTcellmigrationintothepleuralcavityofmiceduringdiverseinflammatoryresponsesisdependentonLTB4/BLT1...
