ThedifferentiationofBcellsintoIg‐secretingplasmacellsrequirestheexpansionofsecretoryorganellestocopewiththeincreasedcargoload.Toevaluatethetimelineofthisprocess,wehavequantitatedthekineticsofsecretoryorganelleexpansionrelativetoIgsecretionandexaminedregulatorycomponentsofsecretorytransportfollowinginvitroactivationofhumanBlymphocytes.UnstimulatedBcellscontainminimalendomembranes.Afteractivation,ERmembraneinductionappearsastightlypackedsphericalstructuresof0.5–1μmdiameterconcentratedinajuxtanuclearposition.Whenthecellsdifferentiateintoplasmablasts,thereisdramaticcell‐sizeincrease,buttheERremainsconcentratedclosetothenucleusandonlylaterfillstheentirecell.Insharpcontrast,previousstudiesinothercelltypeshavefoundthattheERexpandsinsynchronywithincreasingcellsizeduringinterphase,byextensionofERtubulesunderthePM.Inthisstudy,theGolgiremainsconsistentlyasasinglejuxtanuclearstructurebutlinearlyexpandssixfoldinvolumeduringBcellactivation.Furthermore,followingactivecellproliferation,ERexitsitesproliferaterapidly,increasingalmostfourfoldinnumber,inparallelwithasharpincreaseinIgsecretion.ThesefindingsdemonstratethatthecontroloforganellebiogenesisandexpansioninprimaryhumanBcellsaredifferentiallyregulatedbycargofluxcausedbyIgsynthesis...
