OriginallyshowntopromotethegrowthandactivationofBcells,TSLPisnowknowntohavewide‐rangingimpactsonhematopoieticandnonhematopoieticcelllineages,includingDCs,basophils,eosinophils,mastcells,CD4+,CD8+,andNKTcells,Bcells,andepithelialcells.WhereastheroleofTSLPinthepromotionofTH2responseshasbeenstudiedextensivelyinthecontextoflung‐andskin‐specificallergicdisorders,itisbecomingincreasinglyclearthatTSLPmayimpactmultiplediseasestateswithinmultipleorgansystems,includingtheblockadeofTH1/TH17responsesandthepromotionofcancerandautoimmunity.ThisreviewwillhighlightrecentadvancesintheunderstandingofTSLPsignaltransduction,aswellastheroleofTSLPinallergy,autoimmunity,andcancer.Importantly,theseinsightsintothemultifacetedrolesofTSLPcouldpotentiallyallowfornovel,therapeuticmanipulationsofthesedisorders...
