Csf1rmRNAinadultmiceisexpressedincellsofthemacrophagelineage,andduringdevelopment,itisalsoexpressedfromaseparatepromoterinplacentaltrophoblastcells.Thismousetrophoblastpromotersequenceisconservedacrossspecies,buthumantrophoblastsactuallyinitiatetranscriptionfromaseparatepromoter20kbupstream,whichisnotconservedinrodents.A7.2‐kbfragmentofthemouseCsf1rgenomicDNA,includingthe3.5‐kbpromoter,thefirstcodingexonanddownstreamintron,issufficienttodirectreproducibleposition‐andcopynumber‐independentexpressionofanEGFPreporterinvitroandinvivo.Inthisstudy,wehaveexaminedtheconsequenceofremovalofthe150‐bpfragmentencompassingtheconservedtrophoblastpromoterregioninthecontextofthe7.2‐kbpromoteronreportergeneexpressionintransgenicmice.ThedeletionablatedexpressionintheplacentabutalsoabolishedexpressioninmultinucleatedOCLandreducedexpressioninmacrophages.RT‐PCRanalysesofCsf1rmRNArevealedthatmouseOCLuseanotherpromoterwithinthisregion,distinctfromthatusedinplacentaltrophoblasts,togenerateanalternative5′UTR...
