Theextracellularvesicularcompartmenthasemergedasanovelsystemofintercellularcommunication;however,themechanismsinvolvedinmembranevesiclebiogenesisandsecretionareasyetunclear.Amongimmunecellsreleasingmembranevesicles—mastcellsthatresideneartissuesexposedtotheenvironment—aremastermodulatorsofimmuneresponses.Here,wehaveaddressedtheroleofp66Shc,anovelregulatorofmastcellactivationandhomeostasis,inthedynamicreorganizationoftheactincytoskeletonthatisassociatedwithmorphologicalchangesduringsecretion.Weshowthatp66ShcisrecruitedasacomplexwiththelipidphosphataseSHIP1totheF‐actinskeletonandimpairsantigen‐dependentcorticalF‐actindisassemblyandmembranerufflingthroughtheinhibitionofVavandpaxillinphosphorylation.Wealsoshowthatinadditiontoactingasanegativeregulatorofantigen‐dependentmastcelldegranulation,p66Shclimitsthebasalreleaseofgranulecontentsbyinhibitingmicrovesiclebuddingfromtheplasmamembraneandpiecemealdegranulation.Thesefindingsidentifyp66Shcasacriticalregulatorofactindynamicsinmastcells,providingabasisforunderstandingthemolecularmechanismsinvolvedinvesicle‐mediatedsecretioninthesecells...
