TheroleofNOD2andRIP2ininflammatorydiseasehasbeenparadoxical.Whereasloss‐of‐functionNOD2polymorphismscauseCD,agranulomatousdiseaseofthegastrointestinaltract,gain‐of‐functionmutationscauseEOS—agranulomatousdiseaseprimarilyaffectingtheskin,joints,andeyes.Thus,gain‐of‐functionmutationsandloss‐of‐functionpolymorphismscausegranulomatousinflammatorydisease,onlyindifferentanatomiclocations.ThesituationiscomplicatedfurtherbythefactthatWTNOD2andWTRIP2activityhasbeenimplicatedindiseasessuchasasthma,inflammatoryarthritisandMS.ThisarticlereviewstherolethattheNOD2:RIP2complexplaysininflammatorydisease,withanemphasisontheinhibitionofthissignalingpathwayasanovelpharmaceuticaltargetininflammatorydisease...
