Neutrophil extracellular trap formation is elicited in response to cold physical plasma

Coldphysicalplasmaisanionizedgaswithamultitudeofcomponents,includinghydrogenperoxideandotherreactiveoxygenandnitrogenspecies.Recentstudiessuggestthatexposureofwoundstocoldplasmamayacceleratehealing.Uponwounding,neutrophilsarethefirstlineofdefenseagainstinvadingmicroorganismsbuthavealsobeenidentifiedtoplayaroleindelayedhealing.Inthisstudy,weexaminedhowplasmatreatmentaffectsthefunctionsofperipheralbloodneutrophils.Plasmatreatmentinducedoxidativestress,asassessedbytheoxidationofintracellularfluorescentredoxprobes;reducedmetabolicactivity;butdidnotinduceearlyapoptosis.Neutrophiloxidativeburstwasonlymodestlyaffectedafterplasmatreatment,andthekillingofPseudomonasaeruginosaandStaphylococcusaureuswasnotsignificantlyaffected.Intriguingly,wefoundthatplasmainducedprofoundextracellulartrapformation.Thiswasinhibitedbythepresenceofcatalaseduringplasmatreatmentbutwasnotreplicatedbyaddinganequivalentconcentrationofhydrogenperoxide.Plasma‐inducedneutrophilextracellulartrapformationwasnotdependentontheactivityofmyeloperoxidaseorNADPHoxidase2butseemedtoinvolveshort‐livedmolecules.TheamountofDNAreleaseandthetimecourseafterplasmatreatmentweresimilartothatwiththecommonneutrophilextracellulartrapinducerPMA.Afterneutrophilextracellulartrapshadformed,concentrationsofIL‐8werealsosignificantlyincreasedinsupernatantsofplasma‐treatedneutrophils.BothneutrophilextracellulartrapsandIL‐8releasemayaidantimicrobialactivityandspurinflammationatthewoundsite.Whetherthisaidsorexacerbateswoundhealingneedstobetested...
