Angiogenesisintheaorticringmodelisprecededbyactivationoftheimmunesystemandimpairedbyablationofadventitialmacrophages.TreatmentofaorticcultureswithM‐CSFinducedextensiveperiaorticoutgrowthofCD45+CD68+mononuclearcellswithultrastructuralfeaturesofmacrophagesandDCs.PeriaorticlysisofcollagencausedmanyCD45+CD68+cellstoattachtothebottomoftheculturedish.LiftingthecollagengelsleftbehindpatchesofCD45+CD68+cells,whichfocallyorganizedintobranchingcords.ThesecellsalsoexpressedCD14,CD169,F4/80,andα‐SMAbutnotCD31,vWF,desmin,orCD163.DNAsynthesisstudiesshowedthatM‐CSF‐stimulatedcellswereactivelyproliferating.AorticpatchcellsshowedphagocyticpropertiesandrespondedtoIL‐4andGM‐CSFbyexpressingMHCII,differentiatingintoDCs,andformingmultinucleatedgiantcells.TheyalsostimulatedangiogenesisandVEGFproductioninaorticringcultures.Thisstudydemonstratesthattherataortacontainsadistinctsubsetofimmatureimmunocytescapableofproliferating,differentiatingintomacrophagesandDCs,andstimulatingangiogenesis.IsolationofthesecellsinpatchesfromM‐CSF‐stimulatedaorticringsprovidesareproduciblesystemtostudythebiologyandangiogenicroleoftheresidentimmunesystemoftheaorticwall...
