TheG‐CSFisbestknownforitsactivityinthegenerationandactivationofneutrophils.Inaddition,studiesonG‐CSF−/−orG‐CSFR−/−miceandBMCculturessuggestedaroleofG‐CSFinmacrophagegeneration.However,ourunderstandingontheroleofG‐CSFinmacrophagedevelopmentislimited.Here,usinginvitroBMCmodels,wedemonstratedthatG‐CSFpromotedthegenerationofGr‐1high/F4/80+macrophage‐likecellsinM‐BMCs,likelythroughsuppressingcelldeathandenhancinggenerationofGr‐1high/F4/80+macrophage‐likecells.TheseGr‐1highmacrophage‐likecellsproduced“M2‐like”cytokinesandsurfacemarkersinresponsetoLPSandIL‐4/IL‐13,respectively.AdoptivetransferofEGFP‐expressing(EGFP+)M‐BMCsshowedadominant,gut‐homingphenotype.ThesmallintestinallaminapropriaofG‐CSFR−/−micealsoharboredsignificantlyreducednumbersofGr‐1high/F4/80+macrophagescomparedwiththoseofWTmice,butlevelsofGr‐1+/F4/80−neutrophil‐likecellsweresimilarbetweenthesemice.Collectively,theseresultssuggestanovelfunctionofG‐CSFinthegenerationofgut‐homing,M2‐likemacrophages...
