TNF‐αisapleiotropiccytokineinvolvedintheregulationofvariousbiologicaleffects,includingcellsurvivalandproliferation,celldifferentiation,andcelldeath.Moreover,TNF‐αtriggersproinflammatoryresponses,essentiallythroughitsabilitytopromotetheexpressionofvariousproinflammatorygenes.MostofthebiologicaleffectsinitiatedbyTNF‐αrelyonitsabilitytobindtoandactivateTNF‐R1.Asaconsequence,molecularcomplexesarebeingformed,resultingfromtherecruitmentofmultipleadaptorproteinstotheintracellularTNF‐R1DD.TheadaptorproteinFANconstitutivelybindstoaproximalmembranedomainofTNF‐R1calledNSD.Herein,theroleofFANinTNF‐α‐inducedcellsignalingandbiologicalresponsesisdiscussed...
