α‐Tocopherol induces hematopoietic stem/progenitor cell expansion and ERK1/2‐mediated differentiation

【 摘 要 】

Tocopherolspromoteorinhibitgrowthindifferentcelltypes.Inthehematopoieticsystem,theradioprotectivepropertyoftocopherolsisthoughttoactthroughtheexpansionofprimitivehematopoieticcells.However,themechanismsactivatedbytocopherolsandwhichHPsareaffectedremainpoorlyunderstood.Tobetteraddressthesequestions,miceweretreatedwithα‐tocopherol,anditseffectswereinvestigatedintheBMmicroenvironment.α‐TocopherolinducedincreasedproliferationinHSC/HPcells,leadingtoBMhyperplasia.Inaddition,differentiationtothegranulocytic/monocyticlineagewasenhancedbyα‐tocopheroltreatment.α‐TocopheroltreatmentresultedindecreasedbasalphosphorylationofERK1/2,PKC,andSTAT‐5inHSC/HPcells.Incontrast,α‐tocopherolenhancedERK1/2activationinresponsetoIL‐3stimulationinHSC/HPcellswithoutalteringtheexpressionofIL‐3Rs.Moreover,α‐tocopherol‐induceddifferentiationandERK1/2activationwereabolishedinmicepretreatedwithaMEKinhibitor(PD98059);however,pretreatmentwithPD98059didnotreducetheα‐tocopherol‐mediatedincreaseinHSC/HPcellsbutinstead,furtherenhancedtheirproliferation.Therefore,α‐tocopherolinducesexpansionofHSC/HPcellsbyanonidentifiedintracellularpathwayandgranulocytic/monocyticdifferentiationthroughERK1/2activation...
