Intracellularpathogenshavedevelopeddifferentstrategiestosurvivewithinhostcells.Forexample,thesepathogensmightinterferewiththebiogenesisofphagolysosomes,therebyformingreplicativevacuoles.Althoughthecomplexmechanismsusedbypathogenstohijackthebiogenesisofphagolysosomeshavebeenelucidatedinnaiveleukocytes,theroleofleukocyteactivationinthisprocesshasnotyetbeeninvestigated.Leukocytesareknowntobeactivatedbycytokines,andseveralreportshavedemonstratedthatseveralcytokinesmodulatetheendocyticpathwayandthereby,affectphagosomebiogenesis.Theseobservationsprovidemolecularevidencethatendocytosiscanberegulatedbytheimmuneenvironment.Inthisreview,wehighlighttheeffectofleukocyteactivationbycytokinesontheendocyticpathwayandonphagosomebiogenesis.Webrieflydescribethemechanismofphagolysosomeformationbeforefocusingonthestrategiesusedbytwobacterialpathogens,CoxiellaburnetiiandMycobacteriumtuberculosis,tohijackphagolysosomebiogenesis.Finally,weemphasizetheeffectofleukocyteactivationontheendocyticpathwayandonphagolysosomeformation,whichhasnotbeenhighlightedtodate...
