Patientswith10–30dayspostburninjuryaregreatlysusceptibletoinfections.M1MΦ(IL‐10−IL‐12+MΦ)areessentialcellsinhostantibacterialinnateimmunityagainstMRSAinfections.However,theseeffectorcellsarenoteasilygeneratedinhostswhoarecarriersofM2bMΦ(IL‐12−IL‐10+CCL1+LIGHT+MΦ).M2bMΦareinhibitoryonM1MΦgeneration.Inthisstudy,theantibacterialresistanceofmice,10–30dayspostburninjuryagainstMRSAinfection,wasimprovedbythemodulationofM2bMΦactivities.UnburnedmiceinoculatedwithMΦpreparationsfrommice,10–30daysafterburninjury,weresusceptibletoMRSAinfection,whereasunburnedmice,inoculatedwithMΦpreparationsfromthesamemicethatwerepreviouslytreatedwithCCL1antisenseODN,wereresistanttotheinfection.M2bMΦ,isolatedfromDay15burnmice,losttheirM2bMΦproperties3daysaftercultivationunderfrequentmediumchanges,whereastheirM2bMΦpropertiesremainedinthesameculturessupplementedwithrCCL1.Incultures,MΦpreparationsfromDay15burnmicetreatedwithCCL1antisenseODNdidnotproduceCCL1anddidconverttoM1MΦafterheat‐killedMRSAstimulation.Also,Day15burnmicetreatedwiththeODNbecameresistantagainstMRSAinfection.TheseresultsindicatethatCCL1releasedfromM2bMΦisessentiallyrequiredforthemaintenanceoftheirproperties.Theincreasedsusceptibilityofmice,10–30daysafterburninjurytoMRSAinfection,maybecontrolledthroughtheinterventionofCCL1productionbyM2bMΦappearinginassociationwithsevereburninjuries...
