Directedmigrationandarrestofleukocytesduringhomeostasis,inflammation,andtumordevelopmentismediatedbythechemokinesystem,whichgovernsleukocytemigrationandactivities.Althoughweunderstandwelltheeffectsofdifferentchemokinesonebyone,muchlesswasknownaboutthepotentialconsequencesoftheconcomitantexpressionofmultiplechemokinesoroftheirinteractionwithinflammatorymoleculesonleukocytemigrationandfunctions.Inthepast10yr,severalstudiesrevealedtheexistenceofadditionalfeaturesofchemokines:theycanantagonizechemokinereceptorsorsynergizewithotherchemokines,alsobyformingheterocomplexes.Moreover,recentdatashowthatnotonlychemokinesbutalsothealarminhigh‐mobilitygroupbox1canforacomplexwithCXCL12,enhancingitspotencyonCXCR4.Themolecularmechanismunderlyingtheeffectoftheheterocomplexhasbeenpartiallyelucidated,whereasitsstructureisamatterofcurrentinvestigations.ThepresentreviewdiscussesthecurrentknowledgeandrelevanceofthefunctionsofheterocomplexesformedbetweenchemokinesorbetweenthechemokineCXCL12andthealarminhigh‐mobilitygroupbox1.Thesestudieshighlighttheimportanceoftakingintoaccount,whenapproachinginnovativetherapiestargetingthechemokinesystem,alsothefactthatsomechemokinesandmoleculesreleasedininflammation,canconsiderablyaffecttheactivityofchemokinereceptoragonists...
