CirculatingmonocytesexhibitanapoptoticresistancephenotypeduringHIVviremiainassociationwithincreasedMTexpression.MTsareknowntoplayanimportantroleinzincmetabolismandimmunefunction.Wenowshow,inacross‐sectionalstudyusingperipheralmonocytes,thatexpressionofMT1isoformsE,G,H,andXisincreasedsignificantlyincirculatingmonocytecellsfromHIV+subjectsduringchronicviremicepisodesascomparedwithuninfectedsubjects.ThisincreaseinexpressionisalsoobservedduringacuteviremiafollowinginterruptionofsuppressiveART.CirculatingmonocytesfromHIV+donorswerealsofoundtohaveelevatedzincimportergeneZip8expressioninconjunctionwithelevatedintracellularzinclevelsincontrasttoCD4+T‐lymphocytes.InvitroHIV‐1infectionstudieswithelutriatedMDMconfirmadirectrelationbetweenHIV‐1infectionandincreasedMDMMT1(isoformG)geneexpressionandincreasedintracellularzinclevels.Adirectlinkbetweenelevatedzinclevelsandapoptosisresistancewasestablishedusingacell‐permeablezincchelatorTPEN,whichreversedapoptosisresistanceeffectivelyinmonocytesfromHIV‐infectedtolevelscomparablewithuninfectedcontrols.Takentogether,increasesinMTgeneexpressionandintracellularzinclevelsmaycontributedirectlytomaintenanceofanimmune‐activatedmonocytebymediatinganincreasedresistancetoapoptosisduringactiveHIV‐1viremia...
