BTLA,arecentlyclonedcoreceptorexpressedonlymphocytes,negativelyregulatescellactivationbyrecruitingSHP‐1/SHP‐2.However,themechanismsthatregulatetheintracellularlocalizationofBTLAanditstraffickingtothecellsurfaceinTcellsarestillunknown.TodeterminethemechanismsthatregulatetheexpressionofBTLAonthesurfaceofTcells,weexaminedthesubcellularlocalizationofBTLAinmouseTcellsinasteadystate,aswellasuponactivationbyusingaconfocallaser‐scanningmicroscopy.WefoundthatBTLAwaslocalizedmainlyintheGolgiapparatusandsecretorylysosomesinrestingCD4+Tcells.WealsofoundthatintracellularBTLAwastranslocatedtothecellsurfaceandaccumulatedattheimmunologicalsynapseuponTCRstimulation.Furthermore,wefoundthattheBTLA‐HVEMinteractionwasrequiredfortheassociationofBTLAwithlipidrafts.TheseresultsindicatethatthesurfaceexpressionofBTLAanditsaccumulationattheimmunologicalsynapsearetightlyregulatedbyTCRandHVEMstimulationtodeliverefficientinhibitorysignalsintheregulationofCD4+Tcellactivation...
