IL‐2signalingisinvolvedinclonalexpansionofantigen‐specificCD4Tcells.IL‐2isalsoreportedtopromoteTh1butinhibitTh17differentiation,althoughinvivorelevanceremainsunclear.Inaddition,IL‐2‐dependentFoxp3+CD4TregssuppressTcellproliferation,complicatingtheinvivoroleofIL‐2inthedevelopmentofThcellresponses.Toelucidatetherolesofcell‐intrinsicIL‐2signalinginCD4Tcells,wecotransferredTCR‐TgCD4TcellsfromIL‐2Rα(CD25)‐deficientandWTmiceandanalyzeddevelopmentofantigen‐specificTh1andTh17responses.ItwasrevealedthatTh17developmentofantigen‐specificCD4Tcellswaslargelyunaffected,whereasTh1developmentwasimpairedbythelackofIL‐2signaling.SimilardatawereobtainedfrommixedBMchimeraexperimentsusingBMcellsfromCD25‐deficientandWTmice.Inaddition,althoughinvitroblockadeofIL‐2duringTh17developmentgreatlyincreasedthepercentagesofTh17cells,itdidnotaffecttheirnumbers,indicatingthatinvitroTh17developmentisalsoIL‐2‐independent.Th1developmentwasdependentonIL‐2invitroaswell.Thus,ourdatasuggestthatcell‐intrinsicIL‐2signalingiscriticalforTh1developmentbutplaysalimitedroleinTh17developmentinvitroaswellasinvivo...
