InjuryinitiateslocalandsystemichostresponsesandisknowntoincreaseCD4Tregactivityinmiceandhumans.ThisstudyusesaTCRtransgenicTcelladoptivetransferapproachandinvivoTregdepletiontodeterminespecificallytheinvivoinfluenceofTregsonantigen‐drivenCD4Tcellreactivityfollowingburninjuryinmice.WereportherethatinjuryintheabsenceofrecipientanddonorTregspromoteshighantigen‐drivenCD4TcellexpansionandincreasesthelevelofCD4Tcellreactivity.Incontrast,CD4TcellexpansionandreactivityweresuppressedsignificantlyininjuredTreg‐repletemice.Inadditionalexperiments,wefoundthatAPCspreparedfromburn‐orsham‐injured,Treg‐depletedmicedisplayedsignificantlyhigherantigen‐presentingactivitythanAPCspreparedfromnormalmice,suggestingthatTregsmaysuppressinjuryresponsesbycontrollingtheintensityofAPCactivity.Takentogether,thesefindingsdemonstratethatTregscanactivelycontroltheinvivoexpansionandreactivityofantigen‐stimulated,naïveCD4Tcellsfollowingsevereinjury...
