TwomajorsubsetsofhumanMoareidentifiedbasedonCD14andCD16expression:theclassicalCD16–MoandtheminorCD14+CD16+Mo.Invitrostudiessuggesteddistinctfunctionanddifferentiationpotentialforeachcellpopulation.However,theinvivorelevanceofthesefindingsremainsunclear.ToevaluatethedevelopmentandfunctionofhumanMoinaninvivomodel,wetransferredbothMosubpopulationsintotheperitoneumofimmunocompromisedmiceinhomeostaticorinflammatoryconditions.InflammationwasinducedwithsolubleLPSorparticulatezymosan.CD16+weremorephagocyticandproducedhigheramountsofTNFandIL‐6thanCD16–Moearlyaftertransferwithzymosan.Theyalsoproducedhigherlevelsofβ2‐defensininanyconditionevaluated,whichcouldrepresentanewmarkerforthissubpopulation.Incontrast,differentiatingCD16–Mo(24haftertransfer)acquiredgreaterAPCcapacityinLPS‐inducedperitonitis,whereasnoneoftheMosubsetsattainedthisabilitywithzymosan.CX3CL1supportedthesurvivalofbothMosubsetsinvivo.SimilarMosubpopulationswerepresentinhumanperitonitis.TheseresultssupporttheideaofspecializedrolesoftheMosubset,whereCD16+mightactinanimmediateinnateimmuneresponse,whereasCD16–couldhaveamajorroleasAPCs...
