CD4+/CD8+DPthymocytesareawell‐describedTcelldevelopmentalstagewithinthethymus.However,oncedifferentiated,theCD4+lineageortheCD8+lineageisgenerallyconsideredtobefixed.Nevertheless,matureCD4+/CD8+DPTcellshavebeendescribedinthebloodandperipherallymphoidtissuesofnumerousspecies,aswellasinnumerousdiseasesettings,includingcancer.TheexpressionofCD4andCD8isregulatedbyaverystricttranscriptionalprograminvolvingthetranscriptionfactorsRunx3andThPOK.InitiallythoughttobemutuallyexclusivewithinCD4+andCD8+Tcells,CD4+/CD8+Tcellpopulations,outsideofthethymus,haverecentlybeendescribedtoexpressconcurrentlyThPOKandRunx3.ConsiderableheterogeneityexistswithintheCD4+/CD8+DPTcellpool,andthefunctionofCD4+/CD8+Tcellpopulationsremainscontroversial,withconflictingreportsdescribingcytotoxicorsuppressiverolesforthesecells.Inthisreview,wedescribehowtranscriptionalregulation,lineageoforigin,heterogeneityofCD4andCD8expression,age,species,andspecificdiseasesettingsinfluencethefunctionalityofthisrarelystudiedTcellpopulation...
