HistaminemaycontributetothepathologyofMSanditsanimalmodelEAE.WeexploredtheeffectsofhistamineandspecificHRagonistsonactivationandmigratorycapacityofmyelin‐autoreactiveTcells.WeshowthathistamineinvitroinhibitsproliferationandIFN‐γproductionofmouseTcellsactivatedagainstPLP139–151.TheseeffectsweremimickedbytheH1RagonistHTMTandtheH2RagonistdimapritandwereassociatedwithreducedactivationofERK½kinaseandwithincreasedlevelsofcellcycleinhibitorp27Kip‐1,bothinvolvedinTcellproliferationandanergy.H1RandH2Ragonistsreducedspontaneousandchemokine‐inducedadhesionofautoreactiveTcellstoICAM‐1invitroandblockedfirmadhesionofthesecellsininflamedbrainmicrocirculationinvivo.Thushistamine,throughH1RandH2R,inhibitsactivationofmyelin‐autoreactiveTcellsandtheirabilitytotrafficthroughtheinflamedBBB.StrategiesaimedatinterferingwiththehistamineaxismighthaverelevanceinthetherapyofautoimmunediseaseoftheCNS...
