CLRsonDCsplayimportantrolesinimmunityandareexpressedselectivelyoncertainDCsubsets.MurineDCAL2(myeloidinhibitoryC‐typelectin/Clec12a)isatype‐IICLRwithanITIM.UsingamouseDCAL2‐specificmAb,wefoundthatDCAL2isexpressedatrelativelyhighlevelsonAPCsandthatDCAL2expressioncanbeusedtodivideCD8α–DCsintoDCAL2+DCIR2–andDCAL2–DCIR2+subpopulations.CD8α–DCAL2+DC,CD8α–DCIR2+DC,andCD8α+DCAL2+DCsubsetseachexpressdifferentlevelsofTLRsandrespondtouniqueclassesofTLRligandsbyproducingdistinctsetsofcytokines.WhereasCD8α–DCAL2+DCsrobustlyproducecytokines,includingIL‐12,inresponsetoCpG,CD8α–DCIR2+DCsproduceonlyTNF‐αandIL‐10inmodestamountswhenstimulatedwithzymosan.However,CD8α–DCIR2+DCs,unliketheotherDCsubsets,stronglyup‐regulateOX40Lwhenstimulatedwithbacterialflagellin.Aspredictedfromtheircytokineexpression,CD8α–DCAL2+DCsefficientlyinducedTh1responsesinthepresenceofCpGinvitroandinvivo,whereasCD8α–DCIR2+DCsinducedTh2cellsinresponsetoflagellin.Thus,CD8α–DCAL2+DCscompriseadistinctCD8α–DCsubsetcapableofsupportingTh1responses.DCAL2isausefulmarkertoidentifyaTh1‐inducingCD8α–DCpopulation...
