DCsarepotentinitiatorsofadaptiveimmuneresponsestowardinvadingpathogens.Uponreceptionofpathogenicstimuli,DCsinitiateacomplexdifferentiationprogram,culminatinginmatureDCswithanextremecapacitytoactivatenaïveTcells.Duringthismaturation,DCsreducethesynthesisandturnoverofMHCIImolecules.ThisallowsforastablepopulationofMHCII,presentingpeptidescapturedatthetimeandplaceofactivation,thusprovokingspecificimmuneresponsestowardtheactivatingpathogen.TheefficientloadingofantigenicpeptidesontoMHCIIisvitallydependentontheaccessorymoleculeIi,whichaidsintheassemblyoftheMHCIIα‐andβ‐chainsintheERanddirectstheirtraffickingtotheendocyticcompartments,wheretheyencounterendocytosedantigen.However,IiplaysadditionalrolesinDCfunctionbyinfluencingmigration,antigenuptake,andprocessing.ToexaminethebiosyntheticbackgroundfordiverseIifunctionsinDCs,weinvestigatedmRNAandproteinlevelsofIicomparedwithMHCIIinhumanmoDCsduringmaturationusingvariousstimuli.WefindthattheproductionofIididnotcorrelatewiththatofMHCIIandthatmatureDCsmaintainabundantlevelsofIidespiteareducedproductionofnewMHCII...
