Formyl‐peptidereceptorsareafamilyof7transmembranedomain,Gi‐protein‐coupledreceptorsthatpossessmultiplefunctionsinmanypathophysiologicprocessesbecauseoftheirexpressioninavarietyofcelltypesandtheircapacitytointeractwithavarietyofstructurallydiverse,chemotacticligands.Accumulatingevidencedemonstratesthatformyl‐peptidereceptorsarecriticalmediatorsofmyeloidcelltraffickinginthesequentialchemotaxissignalrelaysinmicrobialinfection,inflammation,andimmuneresponses.Formyl‐peptidereceptorsarealsoinvolvedinthedevelopmentandprogressionofcancer.Inaddition,oneoftheformyl‐peptidereceptorfamilymembers,Fpr2,isexpressedbynormalmouse‐colonepithelialcells,mediatescellresponsestomicrobialchemotacticagonists,participatesinmucosaldevelopmentandrepair,andprotectsagainstinflammation‐associatedtumorigenesis.Thesenoveldiscoveriesgreatlyexpandedthecurrentunderstandingoftheroleofformyl‐peptidereceptorsinhostdefenseandaspotentialmoleculartargetsforthedevelopmentoftherapeutics...
