Inthisreport,theconstructionofafunctional,immunocompetent,full‐thicknessskinequivalent(SE)isdescribed,consistingofanepidermalcompartmentcontainingkeratinocytes,melanocytes,andhumanLCsderivedfromtheMUTZ‐3cellline(MUTZ‐LC)andafibroblast‐populateddermalcompartment.TheCD1a+Langerin+HLA‐DR+MUTZ‐LCspopulatetheentireepidermisatasimilardensitytothatfoundinnativeskin.ExposureoftheSEtosubtoxicconcentrationsoftheallergensNiSO4andresorcinolresultedinLCmigrationoutoftheepidermistowardthefibroblast‐populateddermalcompartment.Asignificantdose‐dependentup‐regulationoftheDCmaturation‐relatedCCR7andIL‐1βtranscriptsandofCD83attheproteinleveluponepidermalexposuretobothallergenswasobserved,indicativeofmaturationandmigrationoftheepidermallyincorporatedLC.Wehavethussuccessfullydevelopedareproducibleandfunctionalfull‐thicknessSEmodelcontainingepidermalMUTZ‐LC.Thismodeloffersanalternativetoanimaltestingforidentifyingpotentialchemicalsensitizersandforskin‐basedvaccinationstrategiesandprovidesauniqueresearchtooltostudyhumanLCbiologyinsituundercontrolledinvitroconditions...
